چکیده
Osteoarthritis is currently one of the most common chronic diseases. As life expectancy increases, its prevalence and incidence are expected to rise. At present, more evidences prove correlation between complement system osteoarthritis (OA). This study aims investigate C5's influence on effect MK801 synovial fibroblasts (OA-SFs). We used IL-1b induce OA-SFs derived from mice obtain OA-SFs. And we performed RT-PCR Western Blot assays evaluate expression levels associated mRNA protein. The alteration MAC cell membrane was evaluated by immunofluorescence assay. related inflammatory factors ELISA experiment. could significantly inhibit (OA) marker factors, such as: attack complex (MAC), tumor necrosis factor-α (TNF-α) matrix metalloproteinase-13 (MMP13). Meanwhile, can C5 (C5) in Immunofluorescence assay showed that inhibited MK801. nucleo-plasmic separation experiment demonstrated activation Nuclear factor-κB (NF-κB) signaling pathway Futhermore, koncking down reversed inhibition factors. These results illustrated two points: first, generation release inflammation through C5; second: NF-κB

Connexin43 enhances the expression of osteoarthritis-associated genes in synovial fibroblasts in culture
BACKGROUND Recent work has shown that the gap junction protein connexin43 (Cx43) is upregulated in cells of the joint during osteoarthritis (OA). Here we examined if the OA-associated increase in Cx43 expression impacts the function of synovial fibroblasts by contributing to the production of catabolic and inflammatory factors that exacerbate joint destruction in arthritic disease. METHODS Us...
متن کاملGene expression and activity of cartilage degrading glycosidases in human rheumatoid arthritis and osteoarthritis synovial fibroblasts
INTRODUCTION Similar to matrix metalloproteinases, glycosidases also play a major role in cartilage degradation. Carbohydrate cleavage products, generated by these latter enzymes, are released from degrading cartilage during arthritis. Some of the cleavage products (such as hyaluronate oligosaccharides) have been shown to bind to Toll-like receptors and provide endogenous danger signals, while ...
متن کاملVIP and CRF reduce ADAMTS expression and function in osteoarthritis synovial fibroblasts
ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family is known to play an important role in the pathogenesis of osteoarthritis (OA), working on aggrecan degradation or altering the integrity of extracellular matrix (ECM). Thus, the main purpose of our study was to define the role of vasoactive intestinal peptide (VIP) and corticotrophin-releasing factor (CRF), as immuno...
متن کاملOsteoarthritis and the Complement Cascade
Accumulating evidence demonstrates that complement activation is involved in the pathogenesis of osteoarthritis (OA). However, the intimate complement regulation and cross talk with other signaling pathways in joint-associated tissues remain incompletely understood. Recent insights are summarized and discussed here, to put together a more comprehensive picture of complement involvement in OA pa...
متن کاملSynovial Neuronal Changes in Knee Joint Osteoarthritis
Purpose: The purpose is to study whether pain and inflammation in knee joint osteoarthritis (OA) are associated with local synovial neuronal changes. Methods: Synovial biopsies were harvested from the medial and lateral knee compartments from OA patients undergoing total joint replacement surgery. All patients had predominant pain at the medial joint compartment. Pain and knee joint function we...
